A novel acidophilic, thermophilic iron and sulfur-oxidizing archaeon isolated from a hot spring of tengchong, yunnan, China BJM
Ding,Jiannan; Zhang,Ruiyong; Yu,Yizun; Jin,Decai; Liang,Changli; Yi,Yang; Zhu,Wei; Xia,Jinlan.
A novel thermoacidophilic iron and sulfur-oxidizing archaeon, strain YN25, was isolated from an in situ enriched acid hot spring sample collected in Yunnan, China. Cells were irregular cocci, about 0.9-1.02 µm×1.0-1.31 µm in the medium containing elemental sulfur and 1.5-2.22 µm×1.8-2.54 µm in ferrous sulfate medium. The ranges of growth and pH were 50-85 (optimum 65) and pH 1.0-6.0 (optimum 1.5-2.5). The acidophile was able to grow heterotrophically on several organic substrates, including various monosaccharides, alcohols and amino acids, though the growth on single substrate required yeast extract as growth factor. Growth occurred under aerobic conditions or via anaerobic respiration using elemental sulfur as terminal electron acceptor. Results of...

A reassessment of the phytogeographic characterization of Temperate Pacific South America RChHN
SANTELICES,BERNABE; MENESES,ISABEL.
In 1980, a study of geographic distribution patterns and geographic affinities of benthic algae of temperate Pacific South America (5-55 ºS) provided a first characterization of these coasts. High endemism and limited floristic exchange with the Tropical Pacific and with islands in the South Pacific at various distances from the continent were two outstanding features of the marine flora. A partial blockage of species exchange partially accounted for a peculiar latitudinal pattern of species richness that increases with increasing latitudes. In contrast, there was reduced species richness in comparison with other climatically equivalent regions that have contact with more effective routes of migration. Over the last two decades, more than 30 taxonomic and...

An alternative method to isolate protease and phospholipase A2 toxins from snake venoms based on partitioning of aqueous two-phase systems J. Venom. Anim. Toxins incl. Trop. Dis.
Gómez,GN; Nerli,BB; Acosta,OC; Picó,GA; Leiva,LCA.
Snake venoms are rich sources of active proteins that have been employed in the diagnosis and treatment of health disorders and antivenom therapy. Developing countries demand fast economical downstream processes for the purification of this biomolecule type without requiring sophisticated equipment. We developed an alternative, simple and easy to scale-up method, able to purify simultaneously protease and phospholipase A2 toxins from Bothrops alternatus venom. It comprises a multiple-step partition procedure with polyethylene-glycol/phosphate aqueous two-phase systems followed by a gel filtration chromatographic step. Two single bands in SDS-polyacrylamide gel electrophoresis and increased proteolytic and phospholipase A2 specific activities evidence the...

Antimicrobial resistance and virulence gene profiles in P. multocida strains isolated from cats BJM
Ferreira,Thais Sebastiana Porfida; Felizardo,Maria Roberta; Gobbi,Debora Dirani Sena de; Moreno,Marina; Moreno,Andrea Micke.
Cats are often described as carriers of Pasteurella multocida in their oral microbiota. This agent is thought to cause pneumonia, conjunctivitis, rhinitis, gingivostomatitis, abscess and osteonecrosis in cats. Human infection with P. multocida has been described in several cases affecting cat owners or after cat bites. In Brazil, the cat population is approximately 21 million animals and is increasing, but there are no studies of the presence of P. multocida in the feline population or of human cases of infection associated with cats. In this study, one hundred and ninety-one healthy cats from owners and shelters in São Paulo State, Brazil, were evaluated for the presence of P. multocida in their oral cavities. Twenty animals were positive for P....

Arcobacter butzleri: first isolation report from chicken carcasses in costa rica BJM
Arias,Maria Laura; Cid,Adriana; Fernandéz,Heriberto.
Arcobacter butzleri isolation from chicken carcasses in Costa Rica is reported for the first time. The isolated strains (P and R) were presumptively identified by their phenotypic characteristics. Definitive identification was made using a multiplex PCR assay for the simultaneous detection and identification of Arcobacter butzleri, Arcobacter cryaerophilus and Arcobacter skirrowii. These first isolations indicate the necessity of further investigation about the prevalence, distribution, ecology and interactions with human beings of this and other Arcobacter species.

Banana Supply Chains in Indonesia and Australia: Surviving Isolation from End markets AgEcon
Singgih, Shinta; Woods, Elizabeth.
Two banana supply chains (BSC), one in Indonesia (BSC1) and one in Australia (BSC2), are described based on case study research conducted in 2002. BSC1 sources bananas from small farmers for distribution through traditional markets in Jakarta. BSC2 includes farmers supplying to both major and independent retailers in Sydney through a wholesale agency with some farmer ownership. Farmers in both chains live in relative isolation from the main markets for their bananas. Prices for their bananas depend on the market prices in the city, upon which they have little control. This paper focuses on how farmers removed from key markets, deal with buyers to receive fair prices. In both chains, most transactions between farmers and their buyers were completed through...

CELL CULTURE ISOLATION OF HEPATITIS E VIRUS GENOTYPE 3 STRAIN OBTAINED FROM HUMAN FECES Acta biol.Colomb.
QUINTERO-GIL,Carolina; MIRAZO,Santiago; PARRA-SUESCÚN,Jaime; LÓPEZ-HERRERA,Albeiro; MAINARDI,Victoria; ARBIZA,Juan; ORDÚZ,Sergio.
ABSTRACT Hepatitis E virus (HEV) is considered one of the leading causes of acute viral hepatitis worldwide, and about 20 million infections and approximately 57 000 deaths occurred every year. However, little is known about the replicative virus cycle due to the absence of a consensus cell culture model. A549 cell line is considered susceptible to HEV genotype 3, however, both viral strain and cell culture conditions could affect the viral isolation in vitro. The objective of this work was to isolate in vitro an HEV-3 strain obtained from human feces. To this, a genotype 3 HEV strain previously identified by genetic characterization was inoculated in A549 monolayers, and incubated for two hours at 37 °C. Five days post-infection, cells were passaged...

Characterization of a highly thermostable alkaline phosphatase from the euryarchaeon Pyrococcus abyssi ArchiMer
Zappa, Sebastien; Rolland, Jean-luc; Flament, Didier; Gueguen, Yannick; Boudrant, Joseph; Dietrich, Jacques.
This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Thus far, P. abyssi AP has been demonstrated to be the most thermostable AP, with half-lives at 100 and 105 degreesC of 18 and 5 h, respectively....

Singh,Harkanwaldeep; Rathore,R.S; Singh,Satparkash; Cheema,Pawanjit Singh.
In the present study, the efficacy of polymerase chain reaction (PCR) based on mapA gene of C. jejuni was tested for detection of Campylobacter jejuni in naturally infected as well as spiked faecal and food samples of human and animal origin. Simultaneously, all the samples were subjected to the cultural isolation of organism and biochemical characterization. The positive samples resulted in the amplification of a DNA fragment of size ~589 bp in PCR assay whereas the absence of such amplicon in DNA extracted from E. coli, Listeria, Salmonella and Staphylococcus confirmed the specificity of the primers. Of randomly collected 143 faecal samples comprising human diarrheic stools (43), cattle diarrheic faeces (48) and poultry faecal swabs (52) only 4, 3 and 8,...

Issa,Marina de Azevedo; Soares Filho,Paulo Martins; Fonseca Júnior,Antônio Augusto; Hodon,Mikael Arrais; Santos,Lílian Cristian dos; Reis,Jenner Karlisson Pimenta dos; Cerqueira Leite,Rômulo.
Abstract For the definitive diagnosis of bovine tuberculosis, isolation of the etiologic agent is required. However, there is no consensus on the best methodology for isolation of Mycobacterium bovis in Brazil. This study evaluated the most used decontaminants and culture media in the country, in order to identify the best combination for the Brazilian samples. Three decontaminants - 2% sodium hydroxide (w/v), 0.75% hexadecylpiridinium chloride (w/v) and 5% sulphuric acid (v/v) and four culture media - 7H11 Middlebrook with additives and OADC supplement “A” (7H11 A), the same media with another supplement trademark (7H11 B), tuberculosis blood agar (B83) and Stonebrink's medium were compared. Regarding the isolation, there were no significant differences...

Swine mycobacteriosis is an important cause of carcass condemnation at abattoirs. One of the best ways to recognize the etiologic agent involved, in live animals, is the fecal isolation, as 94% of the lesions are located in the digestive tract. Therefore, the goal of the present study was to compare the performance of four decontamination methods followed by inoculation in three different culture media, totalizing twelve procedures of mycobacteria search from swine fecal samples experimentally contaminated. The swine feces were artificially contaminated with 0.02 g of Mycobacterium avium, PIG-B strain, and subjected to mycobacteria isolation trial. The protocols used were: 1) modified Petroff or basic method; 2) modified Lowenstein-Jensen or acidic method;...

Cultivating the uncultured: limits, advances and future challenges ArchiMer
Alain, Karine; Querellou, Joel.
Since the invention of the Petri dish, there have been continuous efforts to improve efficiency in microbial cultivation. These efforts were devoted to the attainment for diverse growth conditions, simulation of in situ conditions and achievement of high-throughput rates. As a result, prokaryotes catalysing novel redox reactions as well as representatives of abundant, but not-yet cultured taxa, were isolated. Significant insights into microbial physiology have been made by studying the small number of prokaryotes already cultured. However, despite these numerous breakthroughs, microbial cultivation is still a low-throughput process. The main hindrance to cultivation is likely due to the prevailing lack of knowledge on targeted species. In this review, we...

Curtobacterium flaccumfaciens pv. flaccumfaciens detection in bean seeds using a semi-selective culture medium BJM
Maringoni,Antonio Carlos; Camara,Renata de Cassia.
The bacterial wilt caused by Curtobacterium flaccumfaciens pv. flaccumfaciens is currently considered one of the most important bacterial bean disease in Brazil. One of the most effective control methods against this disease is the use of healthy seeds. However, no methods are known that could be routinely used to detect this bacterium in bean seeds under Brazilian condition. The aim of this work was to evaluate qualitative and quantitative detection methods for Curtobacterium flaccumfaciens pv. flaccumfaciens in naturally-infected bean seeds, and the detection of this pathogen in thirty bean seed samples, by sowing onto a semi-selective culture medium the leachate obtained from soaked bean seeds. Both the qualitative and quantitative methods were...
